  • Notes: We previously identified paternal genome heterochromatin (PGH) in male cotton mealybug, demonstrating that two histone methylation events, H3K9me3 and H3K27me3, were enriched in male PGH embryos. To further assess the roles of histone KMTs in embryo development, we employed DAPI staining to detect the appearance of PGH embryos in gravid females at various post-mating times. Using either dsRNA injection or transgenic tobacco expressing dsRNAs, we knocked down two histone KMTs, PsEZH2-X1 and PsEHMT1, and generated evidence that these two KMTs were essential regulators in the male embryo development of cotton mealybug.
