Notes: Genomic DNA was extracted from the scale insect species: Ceroplastes japonicus, Phenacoccus fraxinus, Didesmococcus koreanus and Eulecanium gigantea by using the sodium dodecyl sulfate (SDS) method, the cetyltriethylammonium bromide (CTAB) method, the sodium chloride (NaCl) method, and the acetic acid potassium (KAc) method respectively. Each DNA sample was extracted from the whole body of a single scale insect and was tested by 0.8% agarose gel following electrohporesis. The results suggest that, to some degree, the four methods were useful in extraction of genomic DNA of scale insects. But comparatively, the quality of DNA samples isolated by using the CTAB method and the NaCl method was obviously better and more suitable for PCR than that by using the other two methods.
